Is the 5D II still usable for professional work?

For stills, yes - 21MP full-frame with strong dynamic range still delivers excellent results. For video, it's dated (1080p only, no 4K, no modern AF) but remains a cult camera with distinctive look. Modern AF systems (Dual Pixel, AF tracking) are notably absent.

Can I use EF-S lenses on the 5D II?

No. EF-S lenses have a rear element that protrudes too far into the mirror chamber of full-frame bodies - they will not mount on the 5D II or any other full-frame Canon EF body.
